Howitworks
SimplycloneyourgeneofinterestcontainingaTAG(amber)stopcodonintotheprovidedpEXP4-DEST(Figure1).ThepEXP4-DESTvectorfeaturesattRsitesforefficientGatewayrecombination,C-terminalLumioand6xHistagsforsimplifiedidentificationandpurification,andcomponentsforcell-freeproteinsynthesis.InthepresenceoftheExpresswayTag-On-DemandSuppressortRNA,theTAGcodonisnotrecognizedandtranslationcontinuesthroughtheLumioand6xHistags.IntheabsenceoftheSuppressortRNA,theTAGstopcodonisrecognizedandnativeproteinisproduced(Figure2).Fromthesamevector,youcanrapidlydeterminetheexpressionleveloftheC-terminal-tagged,full-lengthprotein,andthenexpressthenativeproteinfreefromanyadverseaffectsthetagmayhaveonitsstructure,activity,ormolecularinteractions.
